What Is Cell Seeding?
Cell seeding is the process of distributing a planned number of cells into a defined culture vessel at a chosen starting density. The target is commonly expressed as cells per well or cells per unit growth area.

How to Calculate Cells per Well
Multiply the effective cell concentration in cells/mL by the volume seeded into each well in mL.
Cells per well = 1,000,000 × 0.5 = 500,000 cells/well.
How to Calculate the Required Cell Suspension Concentration
Divide the target cells per well by the planned seeding volume per well.
Required concentration = 50,000 ÷ 0.5 = 100,000 cells/mL.
Accounting for Cell Viability
If a measured concentration contains both viable and non-viable cells, a viability correction can be applied when the target is specified as viable cells.
How Much Cell Suspension Is Needed?
For a known effective concentration, calculate the theoretical suspension volume from the total number of target cells:
An optional excess-volume allowance can be applied after the theoretical volume is calculated.
Worked Example: Multiple Wells
Total target = 50,000 × 24 = 1,200,000 viable cells.
Effective concentration = 1,000,000 × 0.90 = 900,000 viable cells/mL.
Required suspension volume = 1,200,000 ÷ 900,000 = 1.333 mL.
Worked Example: Required Concentration
Required concentration = 20,000 ÷ 0.2 = 100,000 cells/mL.
Total target = 20,000 × 96 = 1,920,000 cells.
Cells per Well vs Cells per cm²
Cells per well is useful when the vessel format is fixed. When comparing different culture vessels, seeding density may instead be expressed per unit growth area.
